Synthesis of 1-methyl-3-oxo-7-oxabicyclo[2.2.1]hept-5-ene-2-carboxylic acid methyl ester.
A simple and efficient method for the preparation of 1-methyl-3-oxo-7- oxabicyclo[2.2.1]hept-5-en-2-carboxylic acid methyl ester (1) is described. The first step is a highly regioselective Diels-Alder reaction between 2-methylfuran and methyl-3-bromo- propiolate. A remarkably difficult ketal hydrolysis reaction was effected by treatment with HCl, a simple reagent that was shown to be more effic...
متن کاملStereoselective total synthesis of etnangien and etnangien methyl ester.
A highly stereoselective joint total synthesis of the potent polyketide macrolide antibiotics etnangien and etnangien methyl ester was accomplished by a convergent strategy and proceeds in 23 steps (longest linear sequence). Notable synthetic features include a sequence of highly stereoselective substrate-controlled aldol reactions to set the characteristic assembly of methyl- and hydroxyl-bear...
متن کاملTotal synthesis of (-)-spirangien A and its methyl ester.
The first total synthesis of (-)-spirangien A, a cytotoxic and antifungal polyketide of myxobacterial origin, is reported; this exploits a Stork-Wittig olefination and double Stille cross-coupling sequence to install the sensitive pentaene side chain onto a fully elaborated spiroacetal core, leading initially to the methyl ester of spirangien A.
متن کامل5-(6-Hydroxy-2,5,7,8-tetramethylchroman-2-yl)-2-methyl-pentanoic Acid Methyl Ester
A natural diastereomeric mixture of 5-(6-hydroxy-2,5,7,8-tetramethyl-chroman2-yl)-2-methyl-pentanoic acid methyl ester (1), was isolated from the soft coral Sinularia arborea. The structure of 1 was elucidated by spectroscopic methods and 1 displayed a significantly inhibitory effect on the generation of superoxide anion by human neutrophils.
متن کاملAnti-inflammatory effects of 4-phenyl-3-butenoic acid and 5-(acetylamino)-4-oxo-6-phenyl-2-hexenoic acid methyl ester, potential inhibitors of neuropeptide bioactivation.
Substance P (SP) and calcitonin gene-related peptide (CGRP) are well established mediators of inflammation. Therefore, inhibition of the biosynthesis of these neuropeptides is an attractive potential strategy for pharmacological intervention against a number of inflammatory diseases. The final step in the biosynthesis of SP and CGRP is the conversion of their glycine-extended precursors to the ...
